Glycan Labeling

LudgerTag Glycan Labeling Kit reagents are purified to analytical grade and are dispensed and sealed under clean, inert atmospheres. The ampoules are pre-cleaned by pyrolysis at 500°C then opened just before dispensing and sealing under oxygen-free dry nitrogen. These controls ensure that your analyses work properly each and every time.

Other “instant” labeling systems label the amino group formed during PNGase f release before hydrolysis of the reducing terminal of the sugar. This means these systems cannot be used to label any pre-released glycan standards or other sugars with free reducing ends such as O-glycans, GSL-glycans, or GAGS.

2-AB and 2-AA are used in HILIC-(U)HPLC, WAX-HPLC, and MALDI-MS assays. They are the most referenced glycan label for biopharmaceutical analysis. 2-AA is also used for monosaccharide analysis.

The DMB Sialic Acid Labeling Kit has been developed for the qualitative analysis of sialic acids

Procainamide labeled glycans are commonly used in HILIC-(U)HPLC and ESI-MS assays. Procainamide gives >3 times more response in HPLC assays compared to 2-AB labelled glycans and >30 times more response for ESI-MS.

Permethylation stabilizes sialic acids for MALDI-MS techniques and aids linkage analysis studies (MALDI-MS, ESI-MS).

Procainamide Labeling Kit

Procainamide labeling permits glycan identification by either mass spectrometry or UHPLC. Because of its improved ionisation efficiency compared to 2-AB labelling, it can permit identification of minor glycans (1% relative peak area) by ESI-MS.

This kit is used for the permethylation of glycans. Permethylation stabilizes sialic acids for MALDI mass spectrometry analysis, and aids linkage analysis studies (MALDI-MS, ESI-MS).

2-AB (2-aminobenzamide) is one of the most widely used fluorescent labels for glycosylation analysis. The LudgerTag 2-AB Labeling Kit includes all the reagents required to label up to 30 glycans samples.

Two kits are available which differ in the type of reductant supplied: sodium cyanoborohydride or non-toxic 2-picoline borane.

2-AA (2-aminobenzoic acid) is considered by many to be a superior replacement for 2-AB in most types of complex glycan analysis. 2-AA labeling is reported to have a higher fluorescence and gives higher labelling efficiencies than 2-AB.

The LudgerTag DMB Sialic Acid Labeling Kit has been developed for the qualitative analysis of sialic acids.
The kit contains reagents for the release of sialic acids from glycoproteins. Released sialic acids are conjugated with DMB dye by an amination-cyclisation reaction.

The kit contains reagents and materials for up to 22 samples including the sialic acid reference panel, and the N-acetyl neuraminic acid and N-glycolyl neuraminic acid standards.
